多孔硅纳米颗粒介导外源基因转染

摘 要

多孔硅纳米颗粒因其独特的荧光特性、高比表面积以及良好的生物相容性等,使其在细胞成像、肿瘤成像、生物传感、药物载体等领域具有巨大的应用价值。我们通过电化学腐蚀和超声粉碎的方法制备了具有多孔结构的多孔硅纳米颗粒。再使用聚乙烯亚胺(Polyethylene mine,PEI)辅助,装载增强型绿色荧光蛋白(eGFP),用于体外细胞转染。由于eGFP的荧光强度比GFP大6倍以上,因此其更适合作为一种报告基因来研究基因表达、调控、细胞分化及蛋白质在生物体内定位和转运等。本论文主要致力于使用多孔硅纳米颗粒作为药物载体,介导外源基因(eGFP)细胞内的转染。

eGFP delivery from porous silicon nanoparticles inside cells

ABSTRACT

Porous silicon nanoparticles have been widely applied on the fields of cellular imaging,tumor imaging,biosensors,and drug delivery etc.,because of its unique fluorescence properties,large surface area,high biocompatible. Firstly Porous structure of porous silicon nanoparticles had been fabricated by electrochemical etching and ultrasonic dispersion. Then we use PEI-assisted eGFP loading for in vitro cell transfer. Because eGFP emits fluorescence 6 times greater than GFP. So, eGFP is more suitable as a reporter gene to study gene expression, regulation, cell differentiation and protein localization and transport in vivo. we focused on the use of eGFP delivery from porous silicon nanoparticles inside cells.
